Ga-68 labeled sugar substituted tripodal trisalicylidene imines

Objectives: 68Ga-DOTA labeled compounds are of particular interest for targeted molecular imaging. Metal complexes derived structurally from 1,1,1-tris(aminomethyl)ethane (TAME) are known to complex a variety of metal ions. Tripodal tris(salicylaldimine) from TAME are known to form stable Ga3+ complexes. Here we report the labeling of tripodal tris(salicylaldimine) ligands functionalized with d-galactose 1a or d-glucose 1b with 68Ga as model compounds for the use of radiolabeled sugars for in vivo targeted molecular imaging of carbohydrate and glycobiology pathways.

Methods: A 68Ga generator was eluted with 0.1 M HCl. 68Ga was collected on a SCX cartridge and eluted with 0.4 mL 4.0 M HCl into 0.4 mL 7.0 M HCl. The resulting 5.5 M HCl solution containing 68Ga (as [68GaCl4]-) was collected on an AG1X8 cartridge (30 mg, preconditioned with 2 mL 4.0 M HCl) and after drying (N2), eluted with ca. 0.3 mL of water into a solution of 1 (100 µg) in 100 µL ethanol, 400 µL 0.5 M HEPES buffer and 200 µL 0.6 M NaOH. After adjusting of the pH with 100 µL 0.6 M NaOH to pH 8-9, the reaction mixture was heated at 90°C for 10 minutes to form the final complex.

Results: Sugar substituted imines of type 1a,b form stable 68Ga complexes 2a,b in neutral or basic conditions with a radiochemical purity higher than 95%.

Conclusions: Sugar moieties can be prepared with functional substitutions that enable radiolabeling with 68Ga for targeted molecular imaging applications. The synthesis ligands with varying spacers is subject to further investigation
