Objectives: Generator produced Ga68 is a positron emitting tracer. In the present work both DOTA-substituted octreotide DOTATOC and DOTA-l-NaI3 octreotride were labeled with Ga68 and compared.Ga68 DOTATOC/DOTANOC have been used for neuroendocrine tumors.
Methods: TiO2 based 68Ge/68Ga radionuclide generator (1850-MBq) was obtained from Cyclotron Co. Ltd.Obninsk, Russia. Metal free and analytic reagent grade chemicals, C-18 cartridge, Phenomenex Strata-X Tubes 30 mg and 0.22µm millipore filter were used. 20µg and 40µg of both DOTATOC and DOTANOC were labeled with purified and concentrated 68Ga using suitable concentrations of acetone and hydrochloric acid solutions at 126 oC for 12 min. Labeled peptides were purified on C-18 cartridge and obtained in 400µl ethanol after passing through Millipore filter.
Results: Using 40µg of both the peptides, yield was 50-60%. However, 68Ga-DOTATOC was observed to be more stable in comparison to 68Ga-DOTANOC.Radiochemical purity was found to be >99% for both.
Conclusions: High yields and less time taken during synthesis of injectable 68Ga-labeled somatostatin analog (DOTATOC/DOTANOC) indicate that these are cost effective and time saving agents for evaluating neuroendocrine tumors.
