Objectives: Cyclotron produced 60Cu-ATSM is a hypoxia tracer having demonstrated utility for prognostic assessment of several cancers. PTI has developed a generator/kit produced agent 62Cu-ATSM having the substantial advantage of economical distribution to clinical facilities. Under a corporate IND, PTI is evaluating the safety and biodistribution of this agent through PET imaging in 16 normal volunteers.
Methods: We report the first 3 subjects, all healthy female volunteers injected with 8-17 mCi of 62Cu-ATSM. Each subject was assigned a different bed position: cranial/jaw/neck, mid abdomen, and lower abdominal/pelvis region. Images were collected with high temporal resolution over 45 minutes. ROI analysis was performed for blood pool, brain, gut, kidney, uterus, arm and hip muscles. TACs were fitted with the multi exponential function, y(t)=y0+A1exp(-t/t1)+A2exp(-t/t2)+A3exp(-t/t3).
Results: Values of y0, A1, t1, A2, t2, A3, t3 are as follows. Blood: 5.37E-04, 1.09E-02, 0.19,1.34E-02, 0.19, 7.77E-04; brain: 1.32E-03, 4.49E-03, 1.71, 8.61E-04, 7.59; gut: 5.36E-04, 3.18E-04, 3.70; kidney: 1.12E-03, 3.60E-03, 0.51, 5.48E-04, 25.95; uterus: 1.37E-03, 1.12E-03, 3.05; arm muscle: 1.99E-04, 4.19E-04, 5.93; hip muscle: 7.56E-05, 8.17E-04, 35.33.
Conclusions: Clearance of tracer from the blood pool was rapid. Well perfused organs, brain, kidney, and uterus, showed strong early peak uptake followed by wash out with a half time inversely related to tissue blood flow. Low blood flow tissues such as skeletal muscle rise to a constant value over the first 5 min and remain almost constant out to 40 min. If confirmed in the full group of 16 subjects these measurements could have important ramifications for measurements of hypoxia in tissues including tumors with variable levels blood flow.
