Objectives: 1.To investigation of ENO1 gene function in cancer formation in vitro. 2.Multimodality molecular imaging of human lung cancer animal models with dual reporter genes.
Methods: To generate ENO1-specific monoclonal antibody, we selected anti-ENO1 variant-chain fragments via phage displaying system, and validate the specificity of MAb by flow cytometry and confocal microscopy. Labeling was conducted with 1 µg of anti-ENO1 antibody and 0.9-18 MBq (25-500 µCi) of NH4[131I] using Iodogen method. In vivo targeting was evaluated in MDA-MB-435s/tk-luc and PE089/tk-luc xenografts. Mice were injected with 100-140µCi of 131I-anti-ENO1 antibody via intratumoral or i.v. injection and imaged at 1, 2, 4, and 24 h by gamma scintigraphy.
Results: scFv ENO1 antibody was proved to have high affinity to bind cell-surface ENO1.Significantly high radioactivity uptake of tumor was detected with gamma scintigraphy and whole-body autoradiography,showing that ENO1 could be a good target antigen in human breast cancer.
Conclusions: Anti-ENO1 antibody-conjugated with 131I, as shown in this study, or other radioisotopes have potential clinical applications for human cancer treatment including metastasis.
